Abstract
In order to detect Mycobacterium tuberculosis in cervical lymphadenitis and blood, two-step polymerase chain reaction (PCR) was utilized and IS6110 repeated sequence specific for the bacillus was used as a target for DNA amplification The
reaction
was
processed initially with primer 1 and 2 (5'-ATCCTGCGAGCGTAGCGTAGCGTC-3' and 5'-AGGAGCACATCAGCCGCGTC-3') for 30 cycles followed by nested primer 3 and 4 (5'-CCTGCGAGCGTAGGCGTCGG-3' and 5'-ctcgtccggcgccgcttcgg-3') for 30 cycles.
The results obtained by PCR were compared with the pathological findings or the clinical symptoms; Cervical lymphadenitis tissues of eleven among 13 patients diagnosed as tuberculosis pathologically showed positive PCR results, while 13
lymphadenitis
tissues among 17 patients diagnosed as non-tuberculosis showed negative PCR results. The amplified 123-bps band was seen only in two out of 30 specimens which were prepared from lymph nodes of patients other than lymphadenitis. Bloods of nine
among

pediatric patients diagnosed as tuberculosis clinically or radiologically showed positive PCR results, while bloods of twenty one among 25 podiatric patients diagnosed as non-tuberculosis showed negative PCR results. Bloods of fifteen adult
patients
diagnosed as active tuberculosis showed negative PCR results.
These findings suggest that the tuberculous lymphadenitis tissue is able to be diagnosed by PCR.
